桂枝茯苓胶囊及其成分对LPS诱导RAW264.7细胞产生炎症介质的影响

摘要:

目的 研究桂枝茯苓胶囊及其成分的抗炎活性,并对其抗炎机制进行初步探讨。方法 采用LPS刺激RAW264.7细胞建 立体外炎症模型,用Griess法检测细胞内NO的释放;ELISA法检测桂枝茯苓胶囊及其成分对PGE2、IL-1β、IL-6 和TNF-α分泌的影响;Western Blot法检测桂枝茯苓胶囊及其成分对iNOS和COX-2表达的影响。结果 桂枝茯苓胶囊及其成分能有效抑制LPS刺激下RAW264.7 细胞释放炎症介质NO和PGE2;降低细胞因子IL-1β、IL-6 和TNF-α 的分泌;Western blot 结果表明桂枝茯苓胶囊及其成分能够下调iNOS 和COX-2 蛋白表达水平。结论 桂枝茯苓胶囊及其成分通过抑制iNOS和COX-2的表达进而抑制NO 和PGE2 的分泌,同时抑制炎症因子IL-1β、IL-6 和TNF-α的产生来发挥抗炎作用。

    Influence of releasing inflammatory mediators in the LPS -induced RAW264.7 cells by Guizhi Fuling

    capsule and its ingredients

    ABSTRACT: Objective To study the anti -inflammatory activity of Guizhi Fuling capsule and its ingredients, and to preliminary explore its anti-inflammatory mechanism. Methods Vitro inflammation model was established by using LPS to stimulate RAW264.7 macrophages. Griess reaction was used for assaying the concentration of intra -cellular NO, and ELISA method was performed to measure the expression of PGE2、IL -1β、IL -6 and TNF -α caused by Guizhi Fuling capsule and its ingredients. The expression of iNOS and COX-2 caused by Guizhi Fuling capsule and its ingredients were evaluated by Western Blot. Results Guizhi Fuling capsule and its ingredients remarkably inhibited the release of NO and PGE2, and reduced the secretion of cytokines IL-1 beta, IL-6 and TNF- alpha. Western blot results showed that Guizhi Fuling capsule and its components could down-regulate the expression levels of iNOS and COX-2 protein. Conclusion Guizhi Fuling capsule and its ingredients inhibite NO and PGE2 secretion by inhibiting the expression of iNOS and COX-2, and also inhibited the secretion of IL-1β、IL-6 and TNF-α.
