HDAC6 在小肠癌组织及肠癌细胞中的表达及其下调对肠癌细胞增殖能力的影响

摘要:

目的 研究组蛋白去乙酰化酶6(histone deacetylase 6, HDAC6)在小肠腺癌组织和肠癌细胞中的表达,检测HDAC6低表达对肠癌SW480细胞增殖的影响。方法 应用免疫组织化学法分别检测46例小肠腺癌组织,5例小肠癌旁正常组织和5例小肠正常组织中HDAC6蛋白的表达,采用实时定量RT-PCR和Western blot检测人肠癌细胞株SW480和人肠上皮细胞 株IEC 中HDAC6 mRNA和蛋白的表达,应用脂质体转染siRNA 和CCK-8法检测HDAC6的低表达对SW480细胞增殖能力的影响。结果 小肠腺癌组织中HDAC6蛋白表达的阳性率显著高于小肠癌旁正常组织和小肠正常组织(P<0.05);肠癌 SW480细胞中HDAC6 mRNA和蛋白表达水平显著高于正常IEC细胞(P<0.05)。HDAC6 siRNA下调肠癌SW480 细胞中 HDAC6 mRNA和蛋白的表达。HDAC6表达下调抑制肠癌SW480细胞的增殖。结论 HDAC6 在小肠腺癌组织和肠癌细胞中表 达上调,HDAC6表达下调能抑制肠癌细胞的增殖,提示HDAC6可能与肠癌的发病有关。

    The expression of HDAC6 in intestinal carcinoma and colon cancer cells and the effect of down regulation on the proliferation of colorectal cancer cells

    ABSTRACT: Objective To study the expression of histone deacetylase 6 (HDAC6) in small intestinal adenocarcinoma tissues and colon cancer cells, and to investigate the effect of low expression of HDAC6 on the proliferation of colorectal cancer SW480 cells. Methods Immunohistochemistry was used to detect the expression of HDAC6 protein in 46 patients with small intestinal adenocarcinoma, 5 cases of normal tissue of small intestine carcinoma and 5 cases of normal small intestine tissue. Quantitative RT-PCR and Western blot were performed to detect HDAC6 mRNA and protein expression in human colon cancer cell line SW480 and human intestinal epithelial cell line IEC. Liposomal transfection siRNA and CCK-8 assay were utilized to analyze the effect of HDAC6 siRNA on SW480 cell proliferation. Results The positive rate of HDAC6 protein expression in adenocarcinoma of the small intestine was significantly higher than that in the normal tissue of small intestine carcinoma or normal small intestinal tissue (P<0.05). The expression level of HDAC6 mRNA and protein in SW480 cells were significantly higher than those in IEC cells (P<0.05). HDAC6 siRNA down -regulated the levels of HDAC6 expression and protein in SW480 cells. HDAC6 knockdown inhibited SW 480 cell proliferation. Conclusion HDAC6 is up-regulated in small intestinal adenocarcinoma tissues and colon cancer cells, and downregulation of HDAC6 expression can inhibit the proliferation of colon cancer cells, suggesting that HDAC6 may be involved in the pathogenesis of colorectal cancer.
