宣肺调肝脾汤结合顺尔宁片干预CVA大鼠血清免疫失衡相关细胞因子研究

摘要:

目的 探讨宣肺调肝脾汤结合顺尔宁干预CVA大鼠对血清中免疫失衡相关细胞因子的影响。方法 采用一定比例卵蛋白和氢氧化铝溶液致敏激发的方法建立CVA大鼠模型。32只大鼠随机分为4组,分别为A正常组、B模型组、C顺尔宁片 组、D中药+顺尔宁片组,每组8只,连续给药观察14天。观察大鼠体征变化情况,光镜下观察肺部与气管病变。采用ELISA方法检测各组大鼠血清中IFN-γ 、IL-4的蛋白含量,并进行统计学方法分析。结果 在光镜下图像分析,A组病理变化评分与 B 组比较,具有显著性差异(P<0.05);与B组比较,C组与D组,具有显著性差异(P<0.05);C组与D组比较,差异具有显著 差异(P<0.05);B组血清中IFN-酌蛋白含量与A组比较有明显降低(P<0.01);经过治疗后C 组、D组与模型B组均有明显升 高(P<0.05);C组与D组比较,差异具有统计学意义(P<0.05);与A组比较,B组血清中IL-4蛋白含量明显升高(P<0.01);与模型组B组比较,治疗后C组和D组血清中IL-4蛋白含量值明显降低(P<0.05);C组与D组比较,具有显著性差异(P<0.05)。结论 宣肺调肝脾汤结合顺尔宁片可能通过影响免疫失衡相关细胞因子IFN-γ 、IL-4的蛋白含量,来调节Th1/Th2的免疫失衡,达到防治CVA的作用。

    Study on the cytokines related serum immune imbalance with CVA rats intervened

    by Xuanfei tiaogan-pi decoction and singulair tablets

    ABSTRACT:Objective To Study on the cytokines related serum immune imbalance with CVA rats intervened by Xuanfei tiaoganpi decoction and singulair tablets. Methods The rat model of CVA was established by using a certain percentage of ovalbumin and aluminum hydroxide solution for sensitizing and stimulating the rats. Thirty-two rats were randomly divided into A normal group, B model group, C singulair group, D traditional Chinese medicine combine singulair tablets group, with 8 rats in each group. All the rats were administered continuously for 14 days. The pulmonary and tracheal lesions were observed respectively by naked eye and light microscope. Protein content of IFN-γ , IL-4 cytokines in serum of rats was detected by ELISA, the results were analyzed. Results Under light microscopy image analysis, the difference of the pathological grading in group A and group B showed statistically significant (P<0.05); Compared with group B, C and D group showed statistically significant difference (P<0.05); compared with group D, the difference of group C had significant difference (P<0.05); compared with group A, the IFN -γ  protein content in serum of group B had significantly lower (P<0.01); After treatment, the group C, group D compared with model group B were significantly increased (P <0.05); Group C compared with group D, the difference had statistically significant (P<0.05); compared with group A, the IL- 4 protein content in serum of group B had significant increased (P<0.01); Compared with model group B, the serum IL-4 protein of group C and group D after treatment had significantly decreased (P<0.05); the difference of group C and group D had significant difference (P<0.05). Conclusion Xuanfei tiaogan-pi decoction combined with singulair tablets may regulate immune imbalance of Th1/Th2 by affecting the protein content of IFN-γ , IL-4 cytokines related to the immune imbalance to prevent and cure CVA.
