胶乳增强免疫比浊法与高压液相色谱法测定糖化血红蛋白的方法学比较

摘要:

目的 对胶乳增强免疫比浊法与高压液相层析法(HPLC法)检测糖化血红蛋白(HbA1c)进行方法学比较。方法 收集陕西省渭南市中心医院门诊和住院的患者以及健康体检者的90份临床血液样本,通过糖尿病医学诊疗标准,将待检人员分为: 糖尿病患者组(45例)和非糖尿病患者组(45例),同时采用HPLC 法和胶乳增强免疫比浊法测定HbA1c。对两种方法对两组 受检者的HbA1c测定结果进行精密度、准确度及相关性分析。结果 同一组受检者的两种方法检测结果比较无明显差异(P>0.05);两种方法的批内变异系数(CV)均<2%,批间CV均<4%;r=0.982,回归方程:y=0.133+0.979x。结论 胶乳增强免疫比浊法与HPLC 法均能有效检测HbA1c,且相关性良好,但胶乳增强免疫比浊法检测价格较低,操作简便,可以在中小型医院推广使用。

    Methodology comparison of latex-enhanced immunoturbidimetry and high pressure liquid

    chromatography in the determination of glycosylated hemoglobin

    ABSTRACT: Objective To make the methodology compar ison between latex -enhanced immunoturbidimetry and high pressure liquid chromatography in the determination of glycosylated hemoglobin. Methods Ninety cases of outpatients and inpatient as well as healthy examined people in Weinan Central Hospital were selected, and divided into diabetes patient group (n=45) and non-diabetes patient group (n=45) according to the standard of diabetes diagnosis and treatment, all the patients accepted the HPLC and latex-enhanced immunoturbidimetry for the determination of glycosylated hemoglobin. The precision, accuracy and correlation of the two methods in the two group were compared. Results There was no significant difference between the two methods in the same group (P>0.05). The intra group variation coefficient of the two methods was all below 2%, the inter batch coefficient of variationthe was all below 4%, r=0.982, regression equation: y=0.133+ 0.979x. Conclusion Latex-enhanced immunoturbidimetry and high pressure liquid chromatography all can effectively detect the glycosylated hemoglobin and they have a good correlation, but the latex -enhanced immunoturbidimetry has a lower price, and it is easy to operate, which can be used and promoted in the small and medium-sized hospitals.
