基于Oncomine、TCGA数据库挖掘p57、CKS1及SKP2在肝细胞肝癌中的表达及预后意义

摘要:

目的 通过对Oncomine 及TCGA(The Cancer Genome Atlas)数据库的数据挖掘,探索p57、CKS1及SKP2在肝细胞肝癌 中的表达及预后意义。方法 Oncomine 数据库提取p57、CKS1及SKP2在肝癌(HCC)及癌前和正常组织中转录水平的变化, TCGA数据库下载372例肝癌患者预后及基因表达信息,R3.3.1软件分析p57、CKS1及SKP2与HCC预后的关系。结果 p57在Oncomine数据库大多数HCC队列中呈现转录水平低表达,而在癌前及正常组织高表达;CKS1及SKP2在大多数HCC队列中呈现转录水平的高表达,而癌前及正常组织低表达。p57高表达(5年生存率为50.3%)与低表达(5年生存率为44.5%) 的患者预后没有显著差异(P=0.744)。CKS1低表达(5年生存率为56.0%)患者预后显著好于其高表达(5年生存率为39.2%) 患者(P=0.012)。SKP2高表达(5年生存率为47.9%)与低表达的患者(5年生存率为47%)预后没有显著差异(P=0.380)。结论  p57 mRNA 在HCC 组织中低表达,CKS1及SKP2 mRNA在HCC组织中高表达。CKS1 mRNA表达与HCC预后相关。

    The expression and prognostic significance of p57, CKS1 and SKP2 mRNA in hepatocellular carcinoma utilizing Oncomine and TCGA Datasets

    ABSTRACT: Objective To investigate the expression and prognostic significance of p57, CKS1 and SKP2 mRNA in hepatocellular carcinoma utilizing Oncomine and TCGA datasets. Methods The data about the expression of p57, CKS1 and SKP2 in HCC, precancerous lesions and normal tissues were extracted from Oncomine. Survival and gene expression information from 372 HCC patients were downloaded by TCGA datasets. Survival analysis was performed by R3.3.1 software. Results In most of cohort of Oncomine, the expression of p57 mRNA in HCC was lower than that in precancerous lesions and normal tissues, but the expression of CKS1 and SKP2 mRNA in HCC was higher than that in precancerous lesions and normal tissues. There were no significant differences between patients with low expression of p57 mRNA (5-year survivals of 50.3%) and its high expression (5-year survivals of 44.5%) (P=0.744). The 5-year survival rate in patients with low and high expression of CKS1 were 56.0% and 39.2% respectively, there was significant difference (P=0.012). No significant differences were found in patients with high expression of SKP2 mRNA (5-year survivals of 47.9%) and its low expression (5-year survivals of 47%) (P=0.38). Conclusion p57 mRNA shows high expression and CKS1 and SKP2 mRNA shows low expression in HCC tissue. The expression of CKS1 mRNA has a significant impact on survival of HCC.
